Ultra-Low Temperature Storage: Why Tube Performance Matters at −196 °C

Cryogenic tubes for ultra-low temperature biobanking

Cryogenic storage in biobanking is commonly associated with achieving and maintaining extremely low temperatures. But it is not only that.

When samples are stored down to -196 °C, temperature alone is not the only critical variable. Material behavior, structural integrity and identification systems, all become crucial.

Designed for Ultra-Low Temperatures

Cryogenic tubes are manufactured using thermoplastic polymers such as polypropylene, preferred for their ability to maintain structural stability in low-temperature environments.

As temperatures decrease, material stiffness increases and mechanical behavior changes, making design considerations particularly important.

CryoGen® CLEARLine® tubes by Biosigma are specifically engineered for cryogenic applications.

Manufactured with medical grade polypropylene, they are suitable for biological samples including cells, blood, serum and other fluids at ultra-low temperatures. The closure system, featured with bi-injected caps, helps reduce the risk of fluid loss or contamination.

Closure Integrity Matters

Cap performance plays a crucial role in cryogenic storage. At ultra-low temperatures, the bi-injection cap system is designed  to secure closure and to reduce contamination risk and fluid loss — particularly in workflows involving repeated handling or extended storage periods.

In addition, the availability of color-coded caps supports immediate visual identification, helping to simplify sample organization and reduce handling errors in routine laboratory operations

Traceability Built Into the Tube

In modern biobanking, sample identification is as critical as storage to maintain efficient operations.

All CLEARLine® CryoGen® tubes feature a Code 128 linear barcode on the tube body, supporting standard identification practices in laboratory environments. For operations requiring higher levels of automation and rapid scanning, 2D CryoGen® Tubes are available with a 2D DataMatrix code laser-etched on the bottom.

This dual identification system enhances traceability and reduces reliance on manual data entry, reducing transcription errors. This is particularly Ideal for high-throughput environments.

To support structured inventory management, Biosigma also offers SBS format cryogenic racks, available either empty for operational flexibility or pre-loaded with 2mL and 5mL 2D CryoGen® tubes.

The racks are designed so that the bottom-facing 2D DataMatrix codes remain accessible and readable by automated scanning systems.

The combination of rack organization and dual coding supports:

  • Automated scanning in high-throughput environments
  • Reliable digital inventory tracking
  • Reduced manualtranscription errors

In this way, identification and logistics operate alongside material and closure performance.

Purity and Compliance for Sensitive Applications

Cryogenic storage often involves high-value or irreplaceable biological material. In these contexts, contamination control is not secondary. Tube purity is essential.

CLEARLine® CryoGen® tubes are Sterile (SAL 10⁻⁶) and certified free of human DNA, DNase, RNase, pyrogens, ATP and PCR inhibitors. These specifications support sensitive molecular and cellular applications.

When long-term storage intersects with molecular diagnostics or research workflows, purity and compliance become part of overall system reliability rather than isolated product attributes.

A System Approach to Cryogenic Risk

In biobanking, ultra-low temperature storage is part of a larger system: materials, identification, purity, closure design and sustainability all contribute to long-term reliability.

Selecting cryogenic tubes is not a routine purchasing decision. It is part of a broader strategy to protect sample integrity — today and over time.
